Clonetics™ Intestinal Epithelial Cells

CloneticsTM InEpC's are truly primarycells representing both villi (enterocytes, goblet, and enteroendocrine cells)and crypts structures.  In combinationwith human intestinal myofibroblasts (InMyoFib), InEpC are able to form verytight cell monolayer, representing a unique in vitro system to model humanintestinal homeostasis. Human intestinal epithelial cells (InEpC) are ofpivotal importance in the digestion and absorption of nutrients and theprotection of the organism from bacteria and toxins found in the intestine. Theintestinal epithelium is a highly organized cellular system maintained in adynamic steady state by cells in proliferation and differentiation, which dieand exfoliate into the lumen. Intestinal cells are often difficult to isolate dueto contamination issues and usually have to be cultured and used for assaysimmediately after isolation from tissue.

Cryopreserved Clonetics™ InEpCsare sold at P0, express cytokeratins 8/18 at ≥ 90%,contain 0.8M viable cells / vial and have a seeding efficiency ≥ 50%coverage at certain cell density (see instructions).  Their life span is, post plating, that thecells can be used on days 5-7, and they maintain their properties forapproximately 5 days after that (using Clonetics™ reagents & protocol).  All cells test negative for mycoplasma,bacteria, yeast, and fungi. HIV-1, hepatitis B and hepatitis C are not detectedfor all donors and/or cell lots. A Certificate of Analysis is provided for eachcell lot purchased.  Cells must beincubated at 33 o C!

Clonetics™ Human IntestinalMyofibroblasts

H-InMyoFib share phenotypiccharacteristics of both fibroblasts and smooth muscle cells but do not expresssmooth muscle markers such as smoothelin, caldesmon, or (in the normal colonand small intestine) desmin. They play a fundamental role in growth anddevelopment of intestine, immunology, inflammation, fibrosis, water andelectrolyte transport, contraction and wound healing. They are a useful tool ingeneral gastrointestinal research.

Cryopreserved Clonetics™InMyoFib are guaranteed through 15 population doublings and stain ≥70%postitive for alpha smooth muscle actin and negative (≤10%)for desmin after thaw. They are cryopreserved at passage 2 with ≥500,000cells per vial.  All cells test negativefor mycoplasma, bacteria, yeast, and fungi. HIV-1, hepatitis B and hepatitis Care not detected for all donors and/or cell lots. A Certificate of Analysis isprovided for each cell lot purchased.
