Suc-LLVY-AMC is a sentive fluorogenic substrate for 20S proteasome, calpains and other chymotrypsin-like proteases. The non-fluorescent substrate generates a bright blue fluorescent AMC product that has Ex/Em = 351/430 nm, and can be easily detected with the DAPI filter set. The most common form of the proteasome is known as the 26S proteasome that contains one 20S core particle structure and two 19S regulatory caps. All 20S particles consist of four stacked heptameric ring structures that are themselves composed of two different types of subunits; alpha subunits are structural in nature, whereas beta subunits are predominantly catalytic. The outer two rings in the stack consist of seven alpha subunits each, which serve as docking domains for the regulatory particles and the alpha subunits N-termini form a gate that blocks unregulated access of substrates to the interior cavity. The inner two rings each consist of seven beta subunits and contain the protease active sites that perform the proteolysis reactions.

Cell-Based Assays

At the core of biology is the study of cells - the way they proliferate, the way they function, and ultimately, the way they die. We are particularly excited to introduce our Fluo-8® series, an innovative way to detect Ca2+ in cells. HTS Assays

High-throughput screening, or HTS, has recently become an integral part of biology, particularly in the field of drug discovery. Modern advances in robotics and computer technology have made HTS a more feasible and accessible way to accelerate research. To support this growing usage of HTS, we have designed many of our reagents and kits to be compatible with HTS liquid handling and screening machines. These products have been developed to enable easy and efficient HTS sample generation and data collection. For example, our ultra-sensitive calcium and cAMP GPCR assays feature simplicity in their homogenous and step-saving qualities.

Labeling Probes

Labeling probes allow researchers to determine the presence and quantity of a molecular target. Here at AAT Bioquest, we focus on labeling probes which are intense and stable over time, with quick responses and flexibility over an array of environments. We are especially proud of our line of fluorescent dyes. Our novel iFluor™ series, a line of high quality alternatives to Alexa Fluor®. Not only are iFluor™ dyes available with excitation wavelengths ranging from 350nm to 750nm, but our extensive testing has found that these dyes achieve excellent conjugation yields, produce brighter signals and result in greater sensitivity than similar products on the market.
